Gateway® Technology - Meet the Inventor Series

Learn more at www.lifetechnologies.com This is an interview with Dr Jon Chesnut Life Technologies where he discusses his teams scientific discoveries and inventions that lead to products from Life Technologies. - My name is Jon Chesnut, I'm a Research Fellow in synthetic biology in R&D. At the beginning we were developing cloning vectors, the old pcDNA vectors; it was all restriction enzymes and then we moved into Gateway®, that made cloning that much easier and much more rapid. Tell us about working with Gateway® Technology We started working with Gateway® Technology in 2001, at the time there was standard cloning technology, which is restriction enzyme and ligase, and it was very difficult sometimes to move pieces of DNA from vector to vector. Gateway® Technology set a new paradigm for the customer that allowed them freedom to move pieces of DNA around into different vectors very easy. And once they got clear on the concept the uptake was very quick. And labs were really using Gateway® to move genes and fragments of DNA from one vector to another in a really high- throughput manner. So it really allowed cloning of what we call entire, what we call morphisms, open reading frames from different organisms in very high-throughput from one vector to another; so it really enabled a new paradigm of gene expression tools. What we did further from that is we developed the multi-site Gateway® system which allows assembly of multiple different DNA fragments in a single reaction ...
